Brain activity during walking in older adults: Implications for compensatory versus dysfunctional accounts

activity compared to young adults to perform a given task. This phenomenon has been extensively stud-
ied for cognitive tasks, with the field converging on interpretations described in two alternative accounts.
One account interprets over-activation in older adults as reflecting neural dysfunction (increased brain
activity – indicates poorer performance), whereas another interprets it as neural compensation (in-
creased brain activity - supports better performance). Here we review studies that have recorded brain
activity and walking measurements in older adults, and we categorize their findings as reflecting either
neural dysfunction or neural compensation. Based on this synthesis, we recommend including multiple
task difficulty levels in future work to help differentiate if and when compensation fails as the locomo-
tion task becomes more difficult. Using multiple task difficulty levels with neuroimaging will lead to a
more advanced understanding of how age-related changes in locomotor brain activity fit with existing

Proteome Homeostasis Dysfunction: A Unifying Principle in ALS Pathogenesis
اختلال عملکرد هموستاز پروتئین: یک اصل واحد در پاتوژنز ALS-2020 Amyotrophic lateral sclerosis (ALS) is the most common motor neuron disease
but currently has no effective treatment. Growing evidence suggests that proteome
homeostasis underlies ALS pathogenesis. Protein production, trafficking,
and degradation all shape the proteome. We present a hypothesis that proposes
all genetic lesions associated with ALS (including in mRNA-binding proteins)
cause widespread imbalance to an already metastable proteome. The impact
of such dysfunction is felt across the entire proteome and is not restricted to a
small subset of proteins. Proteome imbalance may cause functional defects,
such as excitability alterations, and eventually cell death. While this idea is a unifying
principle for all of ALS, we propose that stratification will appear that might
dictate the efficacy of therapeutics based on the proteostasis network. |

Reprint of "Executive dysfunction, violence and aggression
چاپ مجدد "اختلال عملکرد اجرایی ، خشونت و پرخاشگری-2020 Executive Functio1ns (EFs) is an umbrella expression used for several processes and subprocesses encompassing
goal-oriented behavior and decision-making strategies. However, the extent to which impaired EFs relate to and
characterize different types of criminal behavior and aggressiveness is not clear. Similarly, research on the
association between EFs and psychopathy has also produced mixed findings. The failure to disaggregate between
different types of crime and antisocial individuals can impede the identification of specific neurocognitive
mechanisms associated with crime. We reviewed the extent to which EFs (according to Miyake latent variable
analysis) are associated with violent and non-violent crime, impulsive and premeditated aggression as well as
psychopathic traits.
Findings, although nonconsensual, suggest a more nuanced view on the association between EFs and crime,
suggesting that different executive domains might be more compromised in violent and impulsive individuals,
and specific psychopathy facets are more associated with executive dysfunction and criminality. Studies in line
with this approach will allow offenders to benefit from intervention strategies that will address their specific
deficits, optimizing their EFs and contributing to better cognitive management in demanding and complex situations,

PRELIMINARY FINDINGS OF THE BRIEF EVERYDAY ACTIVITIES MEASUREMENT (BEAM) IN OLDER ADULTS

subtle declines in instrumental activities of daily living (IADLs) are not always detected in self-reports.
Performance IADL measurements are financially and time burdensome, restricting their use in varied settings.
To address these limitations, we developed the Brief Everyday Activities Measure (BEAM), a short (< 5 minutes)
objective IADL measure that assesses medication and finance management. Design & Participants: The BEAM
was administered to 209 cognitively non-demented community-dwellers (ages 65 – 95 years). Measurements:
Participants completed standardized motor, neuropsychological, psychological, and self-report functional
assessments. Results: BEAM completion time ranged from 54.16 to 259.31 seconds. Interclass correlations
(ICC) for total BEAM completion time was moderate (0.65, 95% CI [.43 -.78]). Accuracy for total BEAM
performance was in the low-moderate range (Kappa = 0.38, p < .001, 95% CI [.18 -.54]). As predicted, lower
accuracy and longer time to complete the BEAM were both associated with worse executive functions, attention,
and processing speed. Conclusions: Medication and finance management can be efficiently assessed within five
minutes. The BEAM may be a valuable screening tool to evaluate these functional abilities.
