1 A self-assembled layer-by-layer surface modification to fabricate the neuron-rich model from neural stem/precursor cells

Background/Purpose: In vitro neural cell-based models have been widely used to mimic the in vivo neural tissue environments and quantitatively understand the effects of pharmaceutical molecules on neural diseases. Recently, several biomimetic neural tissue models have been widely developed by using biomaterials or surface modification. However, the complex protocols of material synthesis or surface modification lack an easy execution to fabricate the neuron favorite environment. Methods: In this study, we utilized a layer-by-layer technique as a surface modification method for regulating the behaviors of neural stem/precursor cells (NSPCs) on material surfaces. Polyelectrolyte multilayers (PEMs) via alternate deposition of poly (allylamine hydrochloride) (PAH) and poly (sodium-4-styrenesulfonate) (PSS) were used to culture NSPCs. After incubation for 7 days, the neuronal differentiation of NSPCs and synapse function of differentiated neurons were identified by immunocytochemistry for lineage specific markers. Results: Compared with the only PAH film, the PSS-ending film (neuron-rich model) was shown to significantly promote differentiation of NSPCs into neurons (more than 50%), form a neuronal network structure; and differentiated neurons exhibiting functional synaptic activity.

2 Comparisons of dual isogenic human iPSC pairs identify functional alterations directly caused by an epilepsy associated SCN1A mutation

Over 1250 mutations in SCN1A, the Nav1.1 voltage-gated sodium channel gene, are associated with seizure disorders including GEFS+. To evaluate how a specific mutation, independent of genetic background, causes seizure activity we generated two pairs of isogenic human iPSC lines by CRISPR/Cas9 gene editing. One pair is a control line from an unaffected sibling, and the mutated control carrying the GEFS+ K1270T SCN1A mutation. The second pair is a GEFS+ patient line with the K1270T mutation, and the corrected patient line. By comparing the electrophysiological properties in inhibitory and excitatory iPSC-derived neurons from these pairs, we found the K1270T mutation causes cell type-specific alterations in sodium current density and evoked firing, resulting in hyperactive neural networks. We also identified differences associated with genetic background and interaction between the mutation and genetic background. Comparisons within and between dual pairs of isogenic iPSC-derived neuronal cultures provide a novel platform for evaluating cellular mechanisms underlying a disease phenotype and for developing patient-specific anti-seizure therapies.

3 In Vitro Differentiated Human Stem Cell-Derived Neurons Reproduce Synaptic Synchronicity Arising during Neurodevelopment

Neurons differentiated from induced pluripotent stem cells (iPSCs) typically show regular spiking and synaptic activity but lack more complex network activity critical for brain development, such as periodic depolarizations including simultaneous involvement of glutamatergic and GABAergic neurotransmission.We generated human iPSC-derived neurons exhibiting spontaneous oscillatory activity after cultivation of up to 6 months, which resembles early oscillations observed in rodent neurons. This behavior was found in neurons generated using a more ‘‘native’’ embryoid body protocol, in contrast to a ‘‘fast’’ protocol based on NGN2 overexpression. A comparison with published data indicates that EB-derived neurons reach the maturity of neurons of the third trimester and NGN2-derived neurons of the second trimester of human gestation. Co-culturing NGN2-derived neurons with astrocytes only led to a partial compensation and did not reliably induce complex network activity. Our data will help selection of the appropriate iPSC differentiation assay to address specific questions related to neurodevelopmental disorders

4 Plasticity of Synaptic Transmission in Human Stem Cell-Derived Neural Networks

Long-term potentiation and depression, inferred from analysis on brain slices, are considered the cellular processes underlying learning and memory formation. They have not so far been demonstrated in human stem cell-derived neurons. By expressing channelrhodopsin in hESCs-derived glutamate neurons and co-culturing them with GABA neurons, we found that blue light stimulation increased the frequency of miniature excitatory postsynaptic currents (mEPSCs) and decreased the ratio of paired pulse facilitation (PPF) in non-ChR2-expressing GABA neurons, indicating a facilitating action at the presynaptic terminals. When paired with postsynaptic depolarization, the repetitive stimulation significantly increased the amplitude of light-evoked EPSCs that persisted during the period, indicating long-term potentiation (LTP). In contrast, low-frequency light stimulation induced long-term depression (LTD). These effects were blocked by N-methyl-D-aspartic acid (NMDA) receptor antagonists, suggesting NMDA receptor-mediated synaptic plasticity in human neural networks. Furthermore, induced pluripotent stem cell (iPSC)-derived neurons of patient with Down syndrome showed absence of LTP or LTD. Thus, our platformoffers a versatile model for assessing human neural plasticity under physiological and pathological conditions

5 HLA concordance between hematopoietic stem cell transplantation patients and umbilical cord blood units: Implications for cord blood banking in admixed populations

Umbilical cord blood stem cell transplantation is an important choice for treating a variety of hematopoietic, neoplastic, and genetic disorders. The optimal size for a cord blood bank to provide matching units for 80% of patients requiring a stem cell transplantation procedure depends on the particular characteristics of each population. In this study, we analyzed the immunogenetic diversity of a sample set of Mexican patients suffering from blood, hematopoietic, and immunological diseases, to assess the best strategy for cord blood banking. For achieving that, we analyzed HLA-A, HLA-B, HLA-DRB1, and HLA-DQB1 genotype and allele frequencies of both units from the bioarchive of the Umbilical Cord Blood Bank from La Raza and patients requiring a stem cell transplant and compared these variables with data from the same geographic and genetic context. We were able to detect significant differences for at least half of the alleles were observed for HLA class I and class II genes between units and patients. Five Native American haplotypes had lower frequencies in patients sample than in the cord blood units. Genetic admixture estimations for both groups showed a higher contribution of Native American component in the cord blood units. Differences in ancestral components in the Umbilical Cord Blood Bank from La Raza and six virtual banks modeled from a pool of Mexican mixed ancestry individuals show that genetic background is important in cord blood collection. In conclusion, increasing diversity over quantity of new cord blood units will improve the cost effectiveness of cord blood banking and health policies regarding hematopoietic stem cell transplantation in admixed populations such as those present in Latin American countries.

6 Evaluating adipocyte differentiation of bone marrow-derived mesenchymal stem cells by a deep learning method for automatic lipid droplet counting

Stem cells are a group of competent cells capable of self-renewal and differentiating into osteogenic, chondrogenic, and adipogenic lineages. These cells provide the possibility of successfully treating patients. During differentiation into adipose tissues, a large number of lipid droplets normally accumulate in these cells, which can be seen through oil red O staining. Although the oil red O staining technique is regularly used for assessing the differentiation degree, its validity for quantitative studies has not been approved yet. Lipid droplet counting has applications in differentiation works and saves time and costs once being automated. In this research, for proving the differentiation of mesenchymal stem cells (MSCs) into adipocyte tissues, their microscopic images were provided. Then, the microscopic images were segmented into square patches, and the lipid droplets were annotated through single-point annotation. The proposed network, based on deep learning, is a fully convolutional regression network processing an image with a small respective field on it. Finally, this method not only does count the lipid droplets but also generates a count map. The average counting accuracy is 94%, which is higher than that of the state-of-the-art methods. It is useful to cell biologists to check the percentage of differentiation in different samples. Also, with a count map, it is possible to observe the regions with high concentrations of lipid droplets without oil red O staining and, thus, examine the total adipocyte differentiation. The contribution of this paper is that a deep learning algorithm has been used for the first time in the field of processing intracellular images.

7 Combined approach for characterization and quality assessment of rabbit bone marrow-derived mesenchymal stem cells intended for gene banking

Rabbit mesenchymal stem cells (rMSCs) are promising agents for the preservation of genetic biodiversity in domestic rabbit breeds. However, rMSCs must meet certain requirements to be used for cryopreservation in animal gene banks. Currently, there are numerous discrepancies in the published data regarding the rMSC phenotype, which may complicate efforts to evaluate their purity and suitability for reuse after cryopreservation in gene and tissue banks. We propose a combined approach (flow cytometry, PCR, differentiation and ultrastructure studies) for the characterization and recovery of rMSCs after cryopreservation. Flow cytometric analyses of rMSCs confirmed the expression of CD29, CD44, vimentin, desmin and α-SMA. RT-PCR revealed the expression of other markers at the mRNA level (SSEA-4, CD73, CD90, CD105, CD146 and CD166). rMSCs showed efficient multilineage differentiation into adipo-, chondro- and osteogenic lineages, SOX2 expression (pluripotency) and typical MSC morphology and ultrastructure. The confirmed rMSCs were subsequently used for cryopreservation. Efficient recovery of rMSCs after cryogenic freezing was demonstrated by high cell viability, normal ultrastructure of reseeded rMSCs, high expression of CD29 and CD44 and lineage differentiation capacity.The proposed combined approach could be used for characterization, cryopreservation and recovery of rMSCs as genetic resources for native rabbit breeds.

با توجه به پیر شدن جمعیت جهان به شکل قابل توجهی آشکار است که نیاز به تولید ایمیلنتها و سیستم عامل ها برای دستکاری رشد سلول های بنیادی در مقیاس دارویی وجود دارد . این مسئله به توجهات سیاسی و اجتماعی بسیاری از کشورها در سراسر جهان نیاز دارد . در این مقاله جزئیات یکی از اولین مطالعات در مورد دستکاری رشد سلولهای بنیادی بر روی سطح لیزر co2 می باشد که به عنوان یک سیستم عامل ارزان دستکاری شد سلولهای بنیادی در مقیاس دارویی شناخته می شود . از طریق درمان لیزری CO2 به طور مجزا تغییراتی در سطوح ایجاد می شود . یکی از این تغییرات این است که زبری سطح نایلون را تا 4.3 میکرومتر افزایش می یابد ، زاویه ی تماس تا 5 درجه مدوله می شود و سطح اکسیژن تا 1% اتم افزایش پیدا می کند . شرح کزانشیمی رشد سلول های بنیادی بر روی نمونه های لیزری ، این است که این نوع از درمان شناخته شود که این مسئله باعث افزایش واکنش ها و در نتیجه باعث افزایش تعدا سلولهای زنده تا 60000 سلول می شود . پارامترهای سطح اصلاح شده توسط درمان سطح لیزری CO2 بر روی واکنش ریشه ی سلول مزانشیم تاثیر می گذارد ، همچنین در این مقاله روند احتمالی که می تواند هدایت واکنش سلولهای بنیادی مزانشیمی شناسایی کندمورد بررسی قرار می گیرد .

کشت سلول های تشکیل دهنده ی استخوان و سلول های اندوتلیال برای تحریک فرایند تشکیل پاتولوژیکی یکی از رگهای خونی یا عروق از استراتژیهایی که برای تلاش های مرتبط سازی بافت استخوانی با عروق یا رگ های خونی موجود ، مورد استفاده قرار می گیرد ، به کار گرفته می شود . در این مطالعه ، سلول های اولیه از بافتهای مختلف برخی حیوانات جدا سازی می شوند ، سلول های بنیادی مغر استخوان موش و اندوتلیال آئورت موش به منظور بررسی اثرات کشت بر روی هم رگ و هم استخوان مورد آزمایش قرار می گیرند . تشکیل لوله ای مانند ساختار 2D که برای اولین بار در مطالعات توسط کشت سلولهای اولیه از همان حیوان مشاهده شد و همچین استخوان ها ور رگها در همان زمان با استفاده از سیستم co-culture مورد بررسی قرار گرفت . سلول های در حال کشت به صورت معدنی شئه اند و تشکیل عروق بسیار ریز از 14 روزگی آنکوباسیون آغاز می شود . با گذشت 28 روز از آنکوباسیون در محیط استخوان ، ترکیب ژن های استخوان در co-cultures به شکل قابل توجهی در مقایسه با تنظیم مثبت RBMSC به تنهایی کشت می شود . این نتایج نشان می دهد که co-cultur از سلولهای آندوتلیال با سلولهای بنیادی مزانشیمی هم استخوان ها و هم رگ ها را تحریک می کنند .
